My understanding of the process is that a package mailed by the USEM must be received by my beneficiary before an interview can be scheduled.  The answer to your question is NO.

So, now that I've waited on the phone for 30 minutes and got my case number.......what do I do with it?

You should get your case file number by email as the letter/email as that is needed for the medical at St. Luke's.   Calling to get your MNL#  from NVC isn't very useful.

Do pay attention to #4 in what to bring to the medical    https://www.visaconnection-philippines.com/medical---slec.html

Didn't I give you this link before?   https://www.visaconnection-philippines.com/us-embassy-usem.html

Plus you should jump over to the Philippines forum for questions regarding the process at the embassy  https://www.visajourney.com/forums/forum/129-philippines/

If you wait for USEM to mail something then you will wait a very long time.   If you know the  case number, go to the embassy website and start following the 5 steps

YMMV

You did send me a link and I've been studying your website ever since.  Very useful.  I read what you said about getting the case number by e-mail and immediately sent NVC an e-mail however I didn't see any harm calling them today since I was stuck by the phone anyway.  I've heard others advocate calling for a number so I thought I'd see what they did with their number.  Hopefully I'll get a e-mail from NVC soon.

Others as in .. in other countries, other embassies, other processes.  Which is why asking your questions in the Philippines forum would get you answers focused  on the process where your fiancee will be processing.

You can setup profile,  prepare ds160, pay the fee,  do CFO, now

Once you get the email from nvc then medical.

Once ceac says ready,  schedule the interview

YMMV

Thought you had read my website ...  all you need is there, including a link to the embassy website and the 5 steps

I love your website.  Pertinent info and easy to read.  However, some of your links don't work.
