My brother was petition by my mother(a greencard) holder in nov 2009, his paper to start visa app and aos arrived, but my brother got married in 2017. So he’s petition is not valid anymore, right? So I just ignore it, or do I need to inform NVC?

You can just ignore it but doesn’t hurt to inform NVC so they close the case for good.

2016-06-30: DS-261 completed, AOS Fee Paid, WL received

There's nothing to approve. The petition became invalid the day they married while their petitioner was an LPR.

They must wait until the parent becomes a USC (if they ever do) then start over with a new petition under F3 category.

The mother may have been eligible for citizenship, but there is nothing in this thread implying that she ever took that step.
