I received the letter of approval from USCIS I-797 and called the NVC and they tell me that the case has not yet come. My question is Can I send an expedite request right now without even having reached the case to the nvc or do I have to wait?

You have to wait for them to receive your file and create your case . Because for now they “don’t know you “ till they enter your informations In their system

The expedite request, after sending the petition, at this point does not make sense and I do not think you'll get one if requested.

You don't need to write on this size of letters, it looks like you are screaming and it's not easy to read the message.
