I am planning to mail out an I-130 Form using USPS Priority Mail Express.

I reside in South Carolina. Do I need to mail my application to the following address?

USCIS
Attn: I-130
131 South Dearborn-3rd Floor
Chicago, IL 60603-5517

Or should I mail it to:

USCIS
P.O. Box 804625
Chicago, IL 60680-4107

Thank you in advanced.

The first address is a physical address for packages sent via UPS, FedEx, etc.......the second address is a post office box for US Postal Service mail.......take your pick.....yes, send it to Chicago.

It is very likely that if you send it USPS to the physical address it will end up in the PO box anyway. At my office we can not receive mail but if someone mails something to us it ends up in the post office box since it is well known to the postal workers of the city. The mail actually has to bounce around a while though because the zip code for my office and the zip code of the PO box are different so the mail is sent to a different post office at first. This is much like the to addresses you have to choose from. So to sum it up I would go with the PO box because best case scenario that get it the day after you send it(they go to the box everyday) worst case scenario they get it the day after you send it. If you send it to the physical address the worst case is it bounces around a couple of days if they do not receive mail at their office. Sorry for the long explanation to a simple question. Just follow the directions they give you and send it to the PO box if using USPS. Everything works better if you follow the directions.

There's "Priority Mail" and there's "Express Mail".  There is no "Priority Express Mail".  I recommend "Priority Mail" sent to the PO Box.  Why try to save a single day in a process that's going to take over a year to complete.  Save your money.
