Which consulate? And where are you? If you're in the US and she's overseas, there may be little chance of her getting a tourist visa with an immediate relative there counting against her.

She can take proof of as many ties as possible to her home country, but bear in mind she may not be asked for them, often the decision is made on the DS-160.

Who's 'the sister'? Not sure how or why she's relevant for your wife's tourist visa?

No, no vaccinations are required for a tourist visa. Yes, letter from employer, proof of property, any family to return too, even things like tickets for concerts or upcoming events. But as above, chances are they won't look at any of those anyway, but take them just in case.

Ah. Does she have a spouse and/or children? If so, she can take proof of them too to show she has to return, if asked.

OK. I'd say chances are slim but she has more chance than if it was your wife (as I originally thought) IMO - as a spouse could adjust status but a sibling can't. So all she can do is try. Good luck.

I agree there is no harm trying.

“If you know the enemy and know yourself, you need not fear the result of a hundred battles. If you know yourself but not the enemy, for every victory gained you will also suffer a defeat. If you know neither the enemy nor yourself, you will succumb in every battle.”

The information they require in the  vast majority of cases is on the application form.

No harm taking anything else but very unlikely to be asked for.

I do remember the NHS had a list of recommended vaccinations depending where you were going, however I do not think there were any for the US.
