I am a Canadian and my spouse American citizen we are in the process of filing for K-1 visa.

I have two children they are over 21 years of age, one is married with children and the other just completed university and working even though they will not be accompanying me to the USA or have any intention of moving to USA. Is it mandatory to include them in the applications?

You need to put them down where it asks you to list all children. That does not mean you are getting them a visa though, it's for record keeping so in 10yrs if you decided you wanted to apply for them it shows you listed them before and they didn't just magically appear.

I am sorry. I am confused. What do you mean your American spouse? If both of you already married, you have to file I-130 for spousal visa, not I-129F for K-1 visa or fiance(e) visa.

Cannot file for a K-1 visa if you have a spouse.

All the children are listed on the I-129F petition where it asks. The petition is not an application for anything, so listing them does not mean they will be applying for a visa.
