Yes, if you want them to be, ensure they both fill out their own I-864 - you will fill one out as well, for a total of 3 - if you decide only one of them will be then:

If a sponsor filed a joint tax return with a spouse, but is qualifying using only his/her own individual income, the sponsor must submit evidence of that individual income. This evidence would include, for example, the sponsor’s own W-2(s), Wage and Tax Statement, and if necessary to reach the income requirement, evidence of other income reported to IRS which can be attributed to him/her, usually on Forms 1099.

No, THEY cannot be joint sponsors. One of them can. Pick the one who has qualifying income and state only their individual income on their I-864. Combining income from multiple sponsors only works when they are part of the same household as the petitioner. The OP indicates they live separately.

Wouldn't the brother fill out an I-864 and if his income/assets is not enough, he'd combine it with his wife who'd fill out an I864A?
