I would wait to submit your packet 3, I waited until the CEAC updated to READY and the file updated twice before submitting the packet 3 contents through email. (You do not need to mail it in) If the embassy has not recieved/logged your file yet, they will just reject the DS-160 and your packet 3 and you will have to do it again. The fees are not paid in packet 3, that is done in Packet 4 when you schedule the interview.

If I remember correctly, no... if by instructions you're thinking of P3, that won't come until your case reaches the consulate. Should just be a couple weeks. 

Nope. It only means that the NVC has received your file. For K visas, just ignore what it says in the box, it's not relevant.

If you got this, does it mean I was suppose to receive something with instruction? Because I didn't O__O"

Yes I also had a Tdap in 2007 at school I think. And got a "booster" from the doctor also. So I may be in same case as you. When I called and asked to verify what vaccination I had they said "tetanus" and nothing else. :O
