It can be anything, they might call your spouse and ask questions to match with your answer, or just simply verify something on your documents.

Timeline

This case is getting a new interview as it was returned to USCIS and the USC had to overcome a NOIR

case returned to embassy in 2019 and now they need to redo medical and interview again

the CO will want to know about the interim and the time spent together to help establish a long term real relationship

the former issues obviously were answered but the CO may ask about them to confirm the evidence given in response to the NOIR

Since we do not know those issues ,  we can't advise on them

but you can tell the VJ community about amount of time the 2 of you have spent together since the 1st interview (a couple of years ago)

and read read read the posts in your embassy and the reviews / that is how to find some of your answers
