My joint sponsor does not have his passport. Would a copy of his birth certificate be acceptable proof of U.S. citizenship?

Yes, it will be accepted. In fact one need not have a passport, if one doesn't desire to go abroad.

If born in the USA, yes. Send a photocopy of a certified copy.

Actually there was no need to presume Citizenship. That was stated. What you presumed was birth in the USA. That's only one of several ways to acquire US Citizenship. Acceptable evidence of US Citizenship include, US Birth Certificates, US Passports, Consular Reports of Birth Abroad, Naturalization Certificates and Certificates of Citizenship. Only when born in the USA is a birth certificate evidence of US Citizenship.

Co-sponsor is not the petitioner so he/she doesn't even need to be US citizen. He/she just need to show he/she paid tax as a resident. So submitting co-sponsor's tax return, which you need to do any way, may be sufficient.

A joint sponsor need not be a US Citizen but must be either a US Citizen or a Legal Permanent Resident of the USA. A tax return is not sufficient. If an LPR instead of Citizen, they submit a photocopy of their green card.
