Hi. My fiance(Sponsor) has Lucas on his birth certificate as his first name,  but Luke on his passport. It was his parents preference. Will this affect the filing to USCIS if he submit different names on passport and birth certificate? Thanks

No.  He doesn't need to submit both documents.   For proof of citizenship he needs to supply one or the other

YMMV

USCIS only requires one document as proof of U.S. citizenship, either send a copy of the birth certificate or passport.     Choose one.

Then use his legal name (on birth certificate) for all documents
