My brother is my cosponsor and has put his current income and his wife's income in Part 6 , question 3 though his income is well above poverty line. His wife is not filing the I864A, but when it comes to recent years tax history, since they both filed jointly, could they use the total amount for each of the last 3 years even though his wife is not filing the I864A?

She needs to file a I-864A if they a) use her income or b) file jointly So in this case, she definitely needs to file one as both conditions apply.
