My fiance will be arriving in the US shortly from Australia, he is on the K1 Visa and already completed a medical examination in Australia. The visa states he cannot enter after September 10, which is about 6 months after his exam.

The USCIS website states the rules were updated in Dec. 2024 and that we must include I-693 when filing Adjustment of Status, I don't believe he got an extra copy of this form from the Australian doctor, so will a new one be required to file for Adjustment of Status or a different form?

Sorry to see you haven't had an answer to this yet.

No, he does not have to file form I-693 with his I-485 adjustment of status form, unless he is missing vaccinations. In that case you need to find a civil surgeon in the US and hope you will find one that will just do the vaccinations, because I have seen a lot of people say that most of them only want to sign off of an I-693 after having done the full medical again. He should have gotten a DS-3025 form (2 pages) from the civil surgeon (aka the vaccination record). If not, request that from them, they have to provide him with that. If the DS-3025 says that the vaccination requirements are completed, that is all he needs to send in with the I-485. See here:

Effective Dec. 2, 2024, you must submit Form I-693, or a partial Form I-693 (such as the Vaccination Record), and you must submit it with your Form I-485. Otherwise, we may reject your Form I-485.

In case his DS-3025 is not complete, there is two options. Either he goes to a civil surgeon in the US that will complete (or redo) the medical with the vaccinations, in which case you send in that signed I-693 with the AOS, or you have a moral or religious conviction against vaccines (as I have) and you send in just the incomplete DS-3025 and an I-601 form to request a waiver with your I-485, plus the fees for this form.

So depends on the situation, but if the DS-3025 states completed, that is all you have to send in. Hope this helps!
