My wife is a citizen and we want to bring her sisters(from Romania) to USA... What to file and how long does it take

Who cares about the population of the us. We have plenty of room for family members....population density is definitely not our problem plus most people that want to come here are good workers.

Obviously you dont understand the rational statement I made. Let me clarify, and break it down for you. If it was easier for family members includling brothers/sisters to come to the U.S. A lot of people will have already made it to the country.

My sister is planning to come to US with a J-1 visa. She is a college student, never married, 21 years old. I'm a US citizen. Can I adjust her status here? Can she get a green card with my help?

No.

It takes 12 years for a USC to petition a sibling - F4 case. Your sister would only be able to adjust if her PD is current. Her PD will not be current for 12 years. When her PD is current, she can adjust for a green card if she is here legally.
