Timeline

I received my case number from NVC a few days ago, but have no idea what to do now. My fiance lives in Santiago, Chile. I don't know should I start the visa application or wait until he receive word from the embassy before doing anything.

This will show when the embassy receives ur file and when it is ready for interview

You can start preparing all the documents you will need for interview, DS160, I-134, etc.

Agreed. Start the ds160 and compile the I-134 support info.

Do not get plane tickets until visa is in hand. We bought plane tickets after we were told another week maybe two. Our tickets were for end of week three. We were moving from friends house to another and one hotel to another and begging consulate to hurry. We also had to spend long time on international phone call with airlines pushing out our flights. It was very very stressful. Do not buy until visa is in hand
