You get on the phone to Knightsbridge EARLY Monday and tell them to hold your medical results until they get a fax from your GP with the MMR details on it. They have to wait on the lab to send back results so it hasn't gone to the embassy if you were just there on .Friday.  They can email you an updated DS-3025 after they have added the shot.

 

If you don't get MMR2 signed off by Knightsbridge before it goes to the embassy, then you take all your records to a USCIS civil surgeon in the US to fill out a new vaccination form (I-693). The kicker is most civil surgeons will not do that unless they give you another complete medical, and TB skin test to the tune of maybe $250 and up. Get it completed by Knightsbridge and you use that form (DS-3025) for AOS.
