good day! My fiance used his affidavit of support twice it was in 2008 (his mom) 2014 (step sister) will I be questioned about his AOS during my interview because he used his aos twice? Please help me. Thanks in advance!

First does your family's income meet the poverty guidelines. http://www.uscis.gov/files/form/i-864p.pdf If so, then no problem.

The amount of times an AOS is submitted has no baring on whether he can move forward with your application for a visa. To apply for a visa, one must supply the necessary documents, which includes financial information via the AOS. So if the answer to the question above is yes, you have NO worries.

The 864, of course, must be current. You just can't resubmit 6 year old documents, if I understand your question.

They are K-1, the I-134 is used, not the I-864

My fiancee and his sister (together with her hubby and son) lives in 1 house but they all have a job, does this mean my fiancee still have to declare them too? My fiancee does not support them financially.

The household size is the number of people a person is responsible for financially. They may or not live in the same house

if your fiancee does not support his sister and her family financially then they are not part of his household size

NO. His sister and her family are not his dependents. He only has to declare his dependents.
