When my wife comes here from PI on her cr-1, then after 2yrs she files for lifting of conditions, do we fill up another I-864 As well as the joint-sponsor?

No, then u wil file the I-751 and follow its instructions which are extremely easy!! http://www.uscis.gov/files/form/I-751.pdf
She can file the I-751 90 days from the date her GREENCARD expires (not the date of ur marriage)

The I-864 is a one time thing, no renewing needed. You file to lift conditions within 90 days of the conditional card expiration.
