If my fiance arrives at JFK on a Saturday or Sunday, can he still get the work visa stamp? I just wondered their hours of operation...

Personally, I think they should be there all the time. Whether you come on weekend or 3 am. So your fiancee should be ok.
My husband got his at 1am Christmas morning, so I'm fairly sure they are always there.
Immigration work 24/7 as long as there are international flights ariving there will be officers available...

O got his at around 8pm on a Tuesday. There was a long line of people and one officer. But he got it, no problem.
