Does it matter which number you use to get updated info from NVC phone line? I'm having trouble keying in NVC number because you must enter first three letters and I am having trouble doing this with my phone.

I haven't tried to dial with uscis numbers, but I think BOTH numbers got 3 letters in front you should dial.
I'm not sure.
I can recommend another way. Download e.i SKYPE buy 10$ credit and you can call from internet using keyboard. You may need also headphones with microphone.

I beleive it is always the uscis number for calling NVC. I dont think there is an option because of all the different embassy's around the world there would not be enough time to list those 3 letters (I have just always used the uscis number to get to the automated system...the NVC case number to talk to an operator)
