During his stay in U.S. my fiance got a DUI...how does this affect his chances of not getting the I-212 approval for the fiance visa?

The document above is the definition of CIMT and what "crimes" qualify. It appears that listed on page 3-5 you'll find that a single DUI conviction isn't a CIMT and therefore wouldn't need a 601 waiver. Just make sure you have all of the documentation about it showing that there were no other offences, etc.
