Can taking unemployment affect spouse visa ?

Unemployment for who and where? If it's for beneficiary in her/his native country, then it doesn't matter. If the petitioner loses the job and goes on unemployment, then there might be concerns about the eligibility due to possible lack of income.

If the process is currently with NVC am i eligible to take unemployment.

It does not affect process and yes you are entitled to unemployment insurance if unemployed. You paid for it.
